چکیده

Introduction Two-Dimensional Gel Electrophoresis technique is a convenient and well-established method to separate thousands of proteins on polyacrylamide gels, according to the differences in their net charge and their molecular mass [1]. Its digital output is an image which depicts proteins as bright or dark spots over a noisy and inhomogeneous background. Each protein is characterized by its vertical and horizontal position on the image, as well as its brightness, size and shape. The process of analyzing 2D-gel images includes the following three main stages [2]: spot detection, spot segmentation and spot quantification. The available software packages [2] for 2D-gel image analysis deal with the problem with a different degree of success and the best solution is far from being reach. In this paper, an original approach for the detection and segmentation of spots on 2D-gel images is presented.

A Novel Spot-Enhancement Anisotropic Diffusion Method for the Improvement of Segmentation in Two-dimensional Gel Electrophoresis Images, Based on the Watershed Transform Algorithm

Introduction Two-dimensional gel electrophoresis (2DGE) is a powerful technique in proteomics for protein separation. In this technique, spot segmentation is an essential stage, which can be challenging due to problems such as overlapping spots, streaks, artifacts and noise. Watershed transform is one of the common methods for image segmentation. Nevertheless, in 2DGE image segmentation, the no...

Segmentation of 2D Gel Electrophoresis Spots Using a Markov Random Field

We propose a statistical model-based approach for the segmentation of fragments of DNA as a first step in the automation of the primarily manual process of comparing two or more images resulting from the Restriction Landmark Genomic Scanning (RLGS) method. These 2D gel electrophoresis images are the product of the separation of DNA into fragments that appear as spots on X-ray films. The goal is...

Unsupervised 2D gel electrophoresis image segmentation based on active contours

This work introduces a novel active contour-based scheme for unsupervised segmentation of protein spots in two-dimensional gel electrophoresis (2D-GE) images. The proposed segmentation scheme is the first to exploit the attractive properties of the active contour formulation in order to cope with crucial issues in 2D-GE image analysis, including the presence of noise, streaks, multiplets and fa...

Automatic protein Spots Quantification in Two-Dimensional gel Images

Two-dimensional (2D) polyacrylamide gel electrophoresis of proteins is a robust and reproducible technique. It is the most widely used separation tool in proteomics. Current efforts in the field are directed at development of tools for expanding the range of proteins accessible with 2D gels. Proteomics was built around the 2D gel. The idea that multiple proteins can be analyzed in parallel grew...

Contour Area Filtering of two-dimensional electrophoresis images

We describe an algorithm, Contour Area Filtering, for separating background from foreground in gray scale images. The algorithm is based on the area contained within gray scale contour lines. It can be viewed as a form of local thresholding, or as a seed growing algorithm, or as a type of watershed segmentation. The most important feature of the algorithm is that it uses object area to determin...
